Persian Abstract

زمینه و هدف: بدخیمی­های معده به عنوان چهارمین سرطان شایع در جهان مطرح می ­باشند. با آن­که بیان بیش از اندازه ژنHER2/neu  در بسیاری از سرطان­ ها با پیشگویی ضعیفی همراه است اما اطلاعات اندکی در رابطه با تکثیر ژنHER2  در سرطان معده با تکنیکMLPA  در دسترس است. مطالعه حاضر با هدف تعیین میزان فراوانی موتاسیون­ های حاصل از مضاعف شدگی ژن HER2/neu با استفاده از تکنیک MLPA در بیماران مبتلا به سرطان معده می­ باشد. روش بررسی: در این مطالعه­ ی توصیفی تعداد 60 نمونه بلاک بافتی بیماران مبتلا به سرطان معده با سابقه اندوسکوپی یا جراحی، بین سال­های 1376 تا 1391 در مرکز بهداشتی–درمانی افضلی پور کرمان از نظر جهش­ های حذف و اضافه در ژن HER2/neu بررسی شدند. نتایج: 73.3 درصد از نمونه‌های مورد مطالعه متعلق به مردان و 26.7 درصد زنان بودند. میانگین سنی نمونه­ های مورد بررسی 62.71 سال بود. به طور کلی فراوانی مضاعف شدگی ژن HER2 در بیماران در سطح پایین 10 درصد و در سطح بالا 3/33 درصد بود.  نتیجه گیری: بطور کلی میزان فراوانی مضاعف شدگی ژن HER2/neu در مردان بیشتر از زنان بوده، با این حال پیشنهاد می ­گردد پژوهش­های بیشتری در این زمینه بعمل آید.

Evaluation of HER2/neu gene amplification frequency in patients with gastric cancer using MLPA method

Abstract

Background and aims:  Gastric malignancies have the fourth place among the most prevalent cancers. In many cancers, overexpressing of HER2/neu gene has been observed with a poor prediction. Up to now, there is a little information about the duplication of HER2/neu gene in gastric cancer using MLPA method. The present study aimed to investigate the frequency of mutations resulting from amplification of HER2/neu gene using MLPA technique in the patients with gastric cancer. Methods: This is a descriptive study which carried out on 60 samples of tissue block obtained from gastric cancer patients with endoscopic and surgical history between 1376-1391 years in Afzalipoor health centers to investigate the deletion and insertion mutations in HER2/neu. Results: The study population consisted of 73.3% females and 26.7% males with the mean age of 62.71. Overall, the frequency of the amplification of HER2/neu gene in the low and high levels was 10% and 3.3%, respectively. Conclusion: It seems that, the amplification frequency of HER2/neu gene in men is more than women. However, it is recommended to conduct further studies.
